Detection of Human Phospho-IKK alpha (S176/S180) by Western Blot.
Western blot shows lysates of HeLa human cervical epithelial carcinoma cell line untreated (-) or treated (+) with 100 nM Calyculin A (Catalog # 1336) and 20 ng/mL Recombinant Human TNF-a (Catalog # 210-TA) for 10 minutes. PVDF membrane was probed with 1 µg/mL of Rat Anti-Human Phospho-IKKa (S176/S180) Mono-clonal Antibody (Catalog # MAB3768) followed by HRP-conjugated Anti-Rat IgG Secondary Antibody (Catalog # HAF005). A specific band was detected for Phospho-IKKa (S176/S180) at approximately 90 kDa (as indicated). Background: IKK alpha
I kappaB kinase alpha (IKK alpha) is also known as IKBKA, CHUK, and IKK1. The classical active IKK complex, composed of IKK alpha, IKK beta, and two forms of processed IKK gamma, phosphorylates and inactivates I kappaB, resulting in the release and nuclear translocation of active NF-kappa B. IKK alpha contains kinase, leucine zipper, and helix-loop-helix domains, and heterodimerizes with IKK beta. NF-kappa B-inducing kinase (NIK) phosphorylates and activates IKK alpha.
