Western blot shows lysates of human kappa light chain purified protein (negative sample), human lambda light chain purified protein, human serum, primate serum, mouse serum (negative sample), and rat serum (negative sample). PVDF membrane was probed with 2 µg/mL of Rabbit Anti-Human/Primate Ig Lambda Light Chain Monoclonal Antibody (Catalog # MAB100492) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog # HAF008). A specific band was detected for Ig Lambda Light Chain at approximately 25 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: Ig Lambda Light Chain

The immunoglobulin light chain is the smaller subunit of an antibody and in humans can be expressed in two types: as a kappa ( kappa) or a lambda ( lambda) chain.  Antibodies are produced by B-Cells that are clonal and each expresses only one type of light chain. The light chain class remains fixed for the life of the B lymphocyte. The ratio of kappa and lambda light chains can be used to determine disease status by immunohistochemistry or ELISA based assays.
