HEK293 human embryonic kidney cell line transfected with human Siglec-16 and eGFP was stained with (A) Mouse Anti-Human Siglec-16 Monoclonal Antibody (Catalog # MAB68192) or (B) Mouse IgG1 Isotype Control Antibody (MAB002) followed by APC-conjugated Anti-Mouse IgG Secondary Antibody (F0101B).Staining was done using our Staining Membrane-associated Proteins protocol.Detection of Siglec-16 in CHO cells transfected with Human Siglec-16 or irrelevant Transfectant cells by Flow Cytometry.
CHO cells transfected with human Siglec-16 (filled histogram) or irrelevant transfectant cells (open histogram) were stained with Mouse Anti-Human Siglec-16 Monoclonal Antibody (Catalog # MAB68192), followed by Allophycocyanin-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # F0101B). To facilitate intracellular staining, cells were fixed and permeabilized with Flow Cytometry Fixation Buffer (Catalog # FC004). Background: Siglec-16
Siglec-16 (Sialic acid-binding Ig-like lectin 16) is a 58-60 kDa member of the CD33-related SIGLEC family of proteins. It is expressed on macrophages and microglia, and based on Siglec-11, likely binds to an alpha2,8-linked sialic acid motif. Although Siglec-16 is assumed to have arisen from a Siglec-11 gene duplication and conversion, it is not an inhibitory receptor but an activating one, and possesses a transmembrane (TM)
