Immunocytochemistry/Immunofluorescence: LAT [p Tyr191] Antibody (2E4) [NBP3-15162] - Immunofluorescence staining of Hela cells with at LAT antibody 1:100, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated Goat Anti-Rabbit IgG (H+L).
Immunohistochemistry: LAT [p Tyr191] Antibody (2E4) [NBP3-15162] - IHC image of LAT antibody diluted at 1:100 and staining in paraffin-embedded human colon cancer. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30 minutes at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

Background: LAT

LAT (linker for activation of T-cells) is involved in the T-cell antigen receptor (TCR) signal transduction pathway, and may play an important role downstream in activating protein tyrosine kinases (1). LAT is phosphorylated by ZAP-70/Syk protein tyrosine kinases leading to recruitment of multiple signaling molecules (2). It has been found that Tyr191 and Tyr171 are required for T-cell activation and Tyr132 is required for the activation of PLCgamma1 and Ras signaling pathways, respectively. Tyr226 and Tyr191 are required for Vav binding, whereas Tyr171 and Tyr132 are necessary for association and activation of phosphoinositide 3-kinase activity and PLCgamma1 (3).
