Background: LXR alpha/NR1H3

LXR (liver X receptors - LXR-alpha and LXR-beta, encoded by NR1H3 and NR1H2, respectively) belongs to nuclear hormone receptor superfamily and are ligand-dependent nuclear transcription factors which are activated by oxysterols. LXR beta is ubiquitously expressed whereas LXR alpha is expressed mainly in liver, intestine, adipose tissue and macrophages. LXR-alpha and LXR-beta shares 78% similarity within their ligand-binding domains and form obligate heterodimers with retinoic X receptor (RXR), bind specific response elements within promoters of target genes ABCA1, ABCG1, SREBF1, APOE etc., and regulate their expression. In the unliganded state, LXRs binds with corepressors such as N-CoR which displaces upon ligand binding. Oxysterols (oxidized cholesterol derivatives) are their natural ligands whereas endogenous ligands includes 22(R)-hydroxycholesterol (22-HC), 24(S)-hydroxycholesterol (24(S)-HC), 24,25-EC , 27-hydroxycholesterol etc. LXRs control the major metabolic pathways such as cholesterol homeostasis, lipogenesis, inflammation, toxicity proliferation cell survival/death and different aspacts of acquired/innate immunity.
