Western blot shows lysates of betaTC-6 mouse beta cell insulinoma cell line. PVDF membrane was probed with 1 µg/mL of Goat Anti-Mouse/Rat PDX-1/IPF1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2517) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF019). A specific band was detected for PDX-1/IPF1 at approximately 45 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 8.

PDX-1/IPF1 was detected in immersion fixed betaTC-6 mouse beta cell insulinoma cell line using Goat Anti-Mouse/Rat PDX-1/IPF1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2517) at 10 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Goat IgG Secondary Antibody (red; Catalog # NL001) and counterstained with DAPI (blue). Simple Western lane view shows lysates of betaTC-6 mouse beta cell insulinoma cell line, loaded at 0.2 mg/mL. A specific band was detected for PDX-1/IPF1 at approximately 48 kDa (as indicated) using 10 µg/mL of Goat Anti-Mouse/Rat PDX-1/IPF1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2517) followed by 1:50 dilution of HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.

Background: PDX-1/IPF1

PDX-1 (Pancreas-duodenum homeobox 1; also IPF-1, IDX-1, STF-1, GSF and IUF-1) is a 42 kDa member of the IPF-1 subfamily, ANTP homeobox family of proteins. It is expressed in developing pancreas, plus pancreatic beta-cells, and duodenal crypt epithelium. In concert with HNF6, PDX-1 drives pancreatic endocrine cell formation by regulating the expression of neurogenin-3. In adult beta-cells, PDX-1 binds to PCIF-1 to regulate insulin production. Mouse PDX-1 is 284 amino acids (aa) in length (Accession # P52946). It contains one transactivation domain (aa 13‑73) a PBX1 P-P-M-W-K dimerization motif (aa 119‑124), and a DNA-binding homeodomain with an embedded NLS (aa 148‑206). Full-length mouse PDX-1 shares 94% and 88% aa identity with rat and human PDX-1, respectively.
