Background: Semaphorin 3B

SEMA3B (semaphorin-3B) belong to semaphorins family, a large class of secreted or membrane-associated proteins which act as chemotactic cues for cell migration and axon guidance during CNS development. Secreted SEMA3s are recruited to plexins by neuropilin (NP) receptors and NP1 preferentially binds to SEMA3B. Neuronal and non-neuronal tissues exhibit abundant and differential expression of SEMA3B which inhibits axonal extension by providing local signals to specify territories inaccessible for growing axons. Besides CNS, semaphorins are expressed in other tissues also wherein they implicate in organogenesis, cell survival, proliferation, apoptosis, migration, angiogenesis, immuno-regulation and tumorigenesis. SEMA3B and another secreted semaphorin SEMA3F have been isolated from a region on human chromosome 3p21.3 which is commonly found deleted in tumors. SEMA3B is either absent or expressed at very low levels in ovarian, lung, and breast cancer cells, and when its expression is restored, it inhibits tumor growth as well as angiogenesis suggesting its role as tumor suppressor.
