Background: Tie-2

The endothelium-specific receptor tyrosine kinase 2 binds and interacts with angiogenic proteins. In acute inflammation, TIE2 signals vascular endothelial cells to stop early leakiness in vessels. This interaction depends partly upon VEGF, PLGF and PDGF. In pregnancy, vascular remodeling is initiated and mediated by VEGF and PLGF; however, recent studies demonstrate that failure of effective vascular remodeling in pre-eclampsia is associated with the erroneously low production of soluble TIE2. Following cerebral stroke, TIE2 production is also critical for appropriate vascular endothelial cell migration and differentiation in response and repair mechanisms. In most cases, damage caused by diabetes, stroke, hypoxia and other cellular insults to vascular endothelial cells is repaired through effective balance of TIE2 with other angiogenic proteins. When the ratio of these proteins is inappropriate, and TIE2 expression decreases, vascular damage occurs.
