This lyophilized preparation is stable at 2-8 degrees C, but should be kept at -20 degrees C for long term storage, preferably desiccated. Upon reconstitution, the preparation is most stable at -20 to -80 degrees C, and can be stored for one week at 2-8 degrees C. For maximal stability, apportion the reconstituted preparation into working aliquots and store at -20 degrees C to -80 degrees C. Avoid repeated freeze/thaw cycles.

Purity

> 98 % pure by SDS-PAGE and HPLC

Endotoxin Level

Less than 0.1 EU/ug of CCL3/MIP-1 alpha as determined by LAL method.

Predicted Molecular Mass

7.9 kDa.
Disclaimer note: The observed molecular weight of the protein may vary from the listed predicted molecular weight due to post translational modifications, post translation cleavages, relative charges, and other experimental factors.

Activity

Ccl3 Lysate is fully biologically active when compared to standard. The biological activity determined by a chemoattract bioassay using human blood monocytes is in a concentration range of 10-100 ng/ml.

Background: CCL3/MIP-1 alpha

CCL3, also known as macrophage inflammatory protein-1alpha (MIP-1alpha), is a cytokine belonging to the CC chemokine family that is involved in the acute inflammatory state in the recruitment and activation of polymorphonuclear leukocytes. Studies showed CCL3 can bind to CCR1, CCR4 and CCR5. MIP-1alpha can induce a dose-dependent inhibition of different strains of HIV-1, HIV-2, and simian immunodeficiency virus (SIV). Recombinant rat MIP-1alpha is a 7.9 kDa protein containing 69 amino acid residues, including the four highly conserved cysteine residues present in CC chemokines and mature rat CCL3 shares 74 %, 91 % and 88 % amino acid sequence identity with human, mouse and cotton rat CCL3, respectively.
