RNAscope™ HiPlex Pro was developed by ACD and Lunaphore to detect up to 12 RNA targets in a method that is optimized for RNA and protein co-detection based on seqIF™ (sequential immunofluorescence) on Lunaphore's hyperplex platform, COMET™. Combining both assays RNAscope HiPlex Pro with seqIF on COMET provides provides end-users with a fully-automated, same-section assay integrating multiplex RNA and multiplex protein data without compromising panel flexibility or sensitivity. The combined workflow allows users to select any 12 RNAscope targets and up to 24 protein targets using off-the-shelf, non-conjugated primary antibodies. The assay is designed to be compatible with current procedures on COMET using a unique protease-free pretreatment to promote streamlined, spatial multiomic data acquisition with subcellular resolution.

Fully-Automated, Protease-Free Workflow: Automated probe hybridization, signal amplification, labeling, imaging and cleaving follow the multiomic-optimized deparaffinization and antigen retrieval steps. Each round allows the detection of 4 RNA targets for up to 3 rounds which can be followed by seqIF protocols.
