Integrin alpha 4 beta 7/LPAM-1: Proteins and Enzymes

Integrin alpha 4 beta 7 (LPAM-1) is an integrin family adhesion receptor that shares subunits with VLA4 and the E-cadherin receptor. It is the non-covalent heterodimer of alpha 4/CD49d and beta 7 transmembrane subunits. Motifs within a4b7 bind to metal ions and regulate cell adhesion functions. a4b7 binds the mucosal addressin MAdCAM-1, as well as VCAM-1 and Fibronectin.

The chemokine receptor CCR9 and a4b7, both critical for homing to the intestinal mucosa, are induced during T cell activation in Peyer’s patches or mesenteric lymph nodes. a4b7-expressing Treg, memory T cells, and B cells home to the intestinal mucosa or the peritoneum. The HIV-1 envelope protein gp120 binds to the active form of a4b7 and may concentrate the virus in gut-associated lymphoid tissue (GALT).
