Recommended Usage

Flow Cytometry

Titration recommended for optimal concentration with starting range of 0.1-1 µg/1 million cells. Sample used for this experiment was HEK 293 cells transfected with eGFP and hPD-1 or irrelevant protein.
Please Note: Optimal dilutions of this antibody should be experimentally determined.

Background: PD-1

Pembrolizumab is a humanized monoclonal IgG4 kappa PD-1 blocking antibody. PD-1 (programmed death receptor-1) is expressed on the surface of lymphocytes, and is also considered an immune checkpoint. The binding of the antibody with PD-1 interferes with the interaction with its ligands PD-L1 and PD-L2 preventing the inhibition of TCR-mediated T-cell proliferation. The inhibition of this PD-1 pathway results in enhanced immunosurveillance and anti-tumor immune response. The original therapeutic has been approved by the FDA for refractory melanoma, as well as other types of cancers such as non-small cell lung cancer, cervical cancer, head and neck cancer and Hodgkin's lymphoma.
