Detection of TNF‑ alpha in Human PBMCs by Flow Cytometry.
Human peripheral blood mononuclear cells (PBMCs) (A) treated with PMA and Ca2+ ionomycin overnight, or (B) resting, were stained with Human Anti-Human TNF- alpha (Research Grade Adalimumab Biosimilar) Monoclonal Antibody (Catalog # MAB9677) followed by Goat anti-Human APC-conjugated Secondary Antibody (Catalog # F0135) and Mouse anti-Human CD3 PE-conjugated Monoclonal Antibody (Catalog # FAB100P). Cytotoxicity Induced by TNF-alpha and Neutralization by Human TNF-alpha Antibody.
Recombinant Human TNF-a (Catalog # 210-TA) induces cytotoxicity in the the L-929 mouse fibroblast cell line in a dose-dependent manner (orange line), as measured by Resazurin (Catalog # AR002). Cytotoxicity elicited by Recombinant Human TNF-a (0.25 ng/mL) is neutralized (green line) by increasing concentrations of Human Anti-Human TNF-a (Research Grade Adalimumab Biosimilar) Monoclonal Antibody (Catalog # MAB9677).
