In a functional flow cytometry test, Recombinant Mouse ICOS Fc Chimera (Catalog # 168-CS, 500 ng/mL) binds to HEK293 human embryonic kidney cell line transfected with recombinant mouse B7-H2 (black dotted line). Binding is completely blocked (orange histogram) by 0.5 µg/mL of Rat Anti-Mouse B7-H2 Monoclonal Antibody (Catalog # MAB10083). Rat IgG2A Isotype Control (Catalog # MAB006) at 0.5 µg/mL was used as a control (blue line). Recombinant Mouse B7-H2 (Catalog # 8127-B7) induces IL-4 secretion in the D10.G4.1 mouse helper T cell line in the presence of Rat Anti-Mouse CD3 Monoclonal Antibody (Catalog # MAB4841) in a dose-dependent manner (orange line), as measured by the Mouse IL-4 Quantikine ELISA Kit (Catalog # M4000B). Under these conditions, IL-4 secretion elicited by B7-H2 (2.5 µg/mL) is neutralized (green line) by increasing concentrations of Rat Anti-Mouse B7-H2 Monoclonal Antibody (Catalog # MAB10083). The ND50 is typically 0.3-1.8 µg/mL.

Neutralization

Background: B7-H2

B7-H2, also called B7RP-1 and ICOSL, is a 60 kDa member of the B7 family of immune costimulatory proteins. It is expressed on resting B cells, dendritic cells, monocytes, alveolar epithelial cells, and placental trophoblasts. B7-H2 interactions with ICOS promote lymphocyte differentiation and activation. Within the ECD, mouse B7-H2 shares 49% and 70% aa sequence identity with human and rat B7-H2, respectively.
