IgG2A

Endotoxin Level

<0.10 EU per 1 μg of the antibody by the LAL method.

Scientific Data Images for Mouse Sonic Hedgehog/Shh N-Terminus Antibody

Alkaline Phosphatase Production Induced by Sonic Hedgehog and Neutralization by Mouse Sonic Hedgehog Antibody.

Alkaline Phosphatase Production Induced by Sonic Hedgehog and Neutralization by Mouse Sonic Hedgehog Antibody.

Recombinant Mouse Sonic Hedgehog N-Terminus (Catalog # 461-SH or 464-SH) induces alkaline phosphatase production in the C3H10T1/2 mouse embryonic fibroblast cell line in a dose-dependent manner (orange line). Alkaline phosphatase production elicited by Recombinant Mouse Sonic Hedgehog N-Terminus (5 µg/mL) is neutralized (green line) by increasing concentrations of Rat Anti-Mouse Sonic Hedgehog N-Terminus Monoclonal Antibody (Catalog # MAB4641). The ND50 is typically 5-15 µg/mL.
Detection of Human and Mouse Sonic Hedgehog/Shh antibody by Western Blot.

Detection of Human and Mouse Sonic Hedgehog/Shh by Western Blot.

Western blot shows lysates of human brain tissue, human stomach tissue, mouse brain tissue, and mouse stomach tissue. PVDF membrane was probed with 2 µg/mL of Rat Anti-Mouse Sonic Hedgehog/Shh N-Terminus Monoclonal Antibody (Catalog # MAB4641) followed by HRP-conjugated Anti-Rat IgG Secondary Antibody (Catalog # HAF005). A specific band was detected for Sonic Hedgehog/Shh at approximately 50 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
Sonic Hedgehog/Shh antibody in Mouse Embryo by Immunohistochemistry (IHC-Fr).

Sonic Hedgehog/Shh in Mouse Embryo.

Sonic Hedgehog/Shh was detected in immersion fixed frozen sections of mouse embryo (11 d.p.c.) using Rat Anti-Mouse Sonic Hedgehog/Shh N-Terminus Monoclonal Antibody (Catalog # MAB4641) at 25 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Rat HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS017) and counterstained with hematoxylin (blue). Specific staining was localized to developing brain. Applications for Mouse Sonic Hedgehog/Shh N-Terminus Antibody

Application
Recommended Usage

Immunohistochemistry

8-25 µg/mL
Sample: Perfusion fixed frozen sections of mouse brain and immersion fixed frozen sections of mouse embryo (11 d.p.c.)

Western Blot

2 µg/mL
Sample: Human brain tissue, human stomach tissue, mouse brain tissue, and mouse stomach tissue

Neutralization

Measured by its ability to neutralize Sonic Hedgehog-induced alkaline phosphatase production in the C3H10T1/2 mouse embryonic fibroblast cell line. The Neutralization Dose (ND50) is typically 5-15 µg/mL in the presence of 5 µg/mL Recombinant Mouse Sonic Hedgehog N-Terminus.

Mouse Sonic Hedgehog Sandwich Immunoassay

ELISA Capture (Matched Antibody Pair)
Recommended Concentration: 2-8 µg/mL
Use in combination with these reagents:
  • Detection Reagent: Mouse Sonic Hedgehog/Shh N-Terminus Biotinylated Antibody (Catalog # BAM4642)
  • Standard: Recombinant Mouse Sonic Hedgehog/Shh, N-Terminus Protein (Catalog # 461-SH)
Please Note: Optimal dilutions of this antibody should be experimentally determined.

Background: Sonic Hedgehog/Shh

Sonic Hedgehog (Shh) is a hedgehog protein that is instrumental in patterning the early embryo. The N-terminal peptide of Shh is released by autoproteolysis and functions through interactions with a multicomponent receptor complex containing the transmembrane proteins Patched and Smoothened.
