Knockdown Validated: STIM1 Antibody [NB110-60547] - Western blot shows lysates of HeLa human cervical epithelial carcinoma parental cell line and Stim-1 knockout (KO) HeLa cell line. PVDF membrane was probed with 1:1000 of Rabbit Anti-Human Stim-1 Polyclonal Antibody (Catalog # NB110-60547) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog #HAF008). Specific band was detected for Stim-1 at approximately 90 kDa (as indicated) in the parental HeLa cell line, but is not detectable in the knockout HeLa cell line. This experiment was conducted under reducing conditions.

Background: STIM1

Stromal interaction molecule 1 (STIM1 or GOK) is a type 1 transmembrane protein that mediates store-operated Ca2+ entry (SOCE), a Ca2+ influx following depletion of intracellular Ca2+ stores. STIM1 is Ca2+ sensor that conveys information about ER lumen's Ca2+ load to store-operated Ca2+ channels. It exists as a single transmembrane protein located in ER membrane with a luminal EF hand and a cytosolic domain, which activates Ca2+ release-activated Ca2+ channels. Upon Ca2+ depletion, STIM1 translocates from ER to plasma membrane where it activates the Ca2+ release-activated Ca2+ (CRAC) channel subunit, TMEM142A/ORAI1 resulting in a strong Ca2+ influx. STIM1 is one of several genes located in imprinted gene domain of 11p15.5, an important tumor-suppressor gene region and alterations in this region have been associated with the Beckwith-Wiedemann syndrome, Wilms tumor, rhabdomyosarcoma, adrenocrotical carcinoma, and lung, ovarian, and breast cancer. STIM1 implicates in early hematopoiesis by mediating attachment to stromal cells and defective STIM1 have been linked to immune dysfunction with T-cell inactivation due to calcium entry defect type 2 (IDTICED2) which is an immune disorder characterized by recurrent infections, impaired T-cell activation and proliferative response, decreased T-cell production of cytokines, lymphadenopathy, and normal lymphocytes counts and serum immunoglobulin levels.
