Enriching High Titer IgG-producing Clones Using Cold-Capture and the Namocell Pala Single Cell Dispenser
Application NotesApplication Notes Summary
Chinese hamster ovary (CHO) cell cloning for recombinant antibodies is vital in monoclonal antibody (mAb) therapeutics. Identifying high-yield clones is challenging due to CHO cell heterogeneity, leading to laborious and time-consuming screening of numerous clones.
The study uses cold-capture to label antibody-producing cells with a fluorescent reagent, allowing sorting based on fluorescence signal for higher productivity cells. They used the Namocell Pala Single Cell Dispenser for single cell isolation. Results showed a 3x increase in IgG production from cold-captured cells compared to non-captured cells.
This method shows promise in improving recombinant antibody production by efficiently identifying high yield clones. Download our app note to learn more about cold-capture-facilitated single cell isolation and cloning's potential benefits.
