Background Information
Enzyme-linked immunospot (ELISpot) assays are scalable, plate-based immunoassays that quantify cytokine secreting cells. These assays are extremely sensitive and can detect 1 cytokine secreting cell in 100,000 cells. Briefly, cytokine secreting cells are incubated in antibody coated wells of a 96 well plate. Secreted analytes are captured by the antibodies. Cells are subsequently washed away and captured analytes are detected with a biotinylated antibody. Analytes are visualized by incubating streptavidin conjugated fluorophore and a fluorescent enhancer or alkaline phosphatase and a chromagen in the wells. Fluorescent or chromogenic spots are then quantified manually or automatically using software.
