Immunohistochemistry: Abeta 40 Antibody (PSH02-82) [NBP3-32077] - Immunohistochemical analysis of paraffin-embedded mouse model of Alzheimer's brain tissue with Rabbit anti-Abeta 40 antibody (NBP3-32077) at 1/1,000 dilution.

The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (NBP3-32077) at 1/1,000 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Background: Abeta 40

Amyloid beta-protein (Abeta) is associated with neuronal injury and death in Alzheimer's disease. The most common isoforms are of Amyloid beta are Abeta40 and Abeta42.

The Amyloid beta A4 protein (Abeta40) is the more common isoform and is the main components of amyloid plaques. Abeta-40 is produced by cleavage of the Amyloid Precursor Protein (APP) that occurs in the endoplasmic reticulum.

The Amyloid beta proteins function as a cell surface receptor and supports neurite growth, neuronal adhesion and axonogenesis on the surface of neurons. Amyloid betas are also involved in cell mobility functions and transcription regulation via various protein-protein interactions.

Abeta40 antibodies are useful tools for Alzheimer's studies and research on amyloid plaques.
