Western Blot: Acetyl-CoA Carboxylase alpha/ACACA [p Ser79] Antibody (JE63-95) [NBP3-33135] - Western blot analysis of Acetyl-CoA Carboxylase alpha/ACACA on different lysates with Rabbit anti-Acetyl-CoA Carboxylase alpha/ACACA antibody (NBP3-33135) at 1/2,000 dilution.

Lane 1: NIH/3T3 cell lysate
Lane 2: NIH/3T3 treated with 0.5uM Oligomycin for 30 minutes cell lysate
Lane 3: NIH/3T3 treated with 0.5uM Oligomycin for 30 minutes then treated with pp for 1 hour cell lysate

Exposure time: 1 minutes 14 seconds;

4-20% SDS-PAGE gel.

Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (NBP3-33135) at 1/2,000 dilution was used in 5% NFDM/TBST at 4C overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1/50,000 dilution was used for 1 hour at room temperature.

Background: Acetyl-CoA Carboxylase alpha/ACACA

Acetyl-CoA carboxylase 1 (ACC1) is a biotin dependent lipogenic enzyme that is highly expressed during adipogenesis. ACC1 catalyzes acetyl-CoA carboxylation, producing malonyl-CoA, a metabolite involved in energy homeostasis regulation. Malonyl-CoA is a two carbon donor in the synthesis of long-chain fatty acids and the elongation of fatty acids found in the cystol (1). ACC1 is regulated short-term by citrate, CoA, and palmitoyl-CoA through allosteric interactions. Nutrients and hormones can be both short-term (inducing reversible phosphorylations by such as AMPK) and long-term (transcription level regulation) regulators of ACC1 (2). Highly expressed in lipogenic tissues, ACC1 is found in liver, adipose, and lactating mammary gland (3). ACC1 has been implicated as a target in the development of anti-obesity drugs (4).
