Western Blot: Adenylate Cyclase 3 Antibody [NBP3-05526] - Western blot analysis of HEK293 cell lysates using Adenylate Cyclase 3 Antibody, dilution 1:1,000, in green: [1] protein standard, [2] non-transfected HEK293 cells, and [3] HEK293 cells transfected with a vector encoding Myc-DDK tagged fill length human adenylate cyclase 3. The strong band at about 130kDa in transfected cells demonstrates overexpression of the human adenylate cyclase 3 protein, and bands over 250kDa presumably correspond to either heavily glycosolated form of adenylate cyclase 3 or multimeric forms of the protein. The same blot was simultaneously probed with rabbit polyclonal antibody to GAPDH dilution 1:20,000 in red, which reveals the single band at ~37kDa seen in both transfected and non-transfected cells.

The Blue is Hoechst stain revealing nuclei. The Adenylate Cyclase 3 Antibody reveals neuronal cilia while the tyrosine hydroxylase antibody reveals the axons of chatecholaminergic neurons.

Background: Adenylate Cyclase 3

ADCY3, also known as Adenylate cyclase type 3, is a 129 kDa 1,144 amino acid protein, and is involved in the formation of the secondary messenger protein, cAMP. This protein has also been shown to have interactions with STOML3, CAMK2G, PDE1A, PDE1C, and PDE2A in the Signaling by FGFR, PKA activation in glucagon signaling, DAG and IP3 signaling, Regulation of Water Balance by Renal Aquaporins, and Opioid Signalling pathway. Disease research is currently being studied with relation to ADCY3 and cholera, alcoholism, tuberculosis, and breast cancer.
