Immunohistochemistry: AGL Antibody (PSH04-56) [NBP3-32009] - Immunohistochemical analysis of paraffin-embedded human liver tissue with Rabbit anti-AGL antibody (NBP3-32009) at 1/1,000 dilution.

The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (NBP3-32009) at 1/1,000 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Lane 1: K-562 cell lysate (20 ug/Lane)
Lane 2: HEK-293 cell lysate (20 ug/Lane)
Lane 3: Mouse skeletal muscle tissue lysate (10 ug/Lane)
Lane 4: Rat skeletal muscle tissue lysate (10 ug/Lane)

Lysates/proteins at 10 ug/Lane.

Predicted band size: 175 kDa
Observed band size: 150 kDa

Exposure time: Lane 1-2: 3 minutes; Lane 3-4: 20 seconds; ECL;

4-20% SDS-PAGE gel.

Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (NBP3-32009) at 1/1,000 dilution was used in 5% NFDM/TBST at 4℃ overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1/50,000 dilution was used for 1 hour at room temperature.

Background: AGL

AGL encodes the glycogen debrancher enzyme which is involved in glycogen degradation. This enzyme has two independent catalytic activities which occur at different sites on the protein: a 4-alpha-glucotransferase activity and a amylo-1,6-glucosidase activity. Mutations in this gene are associated with glycogen storage disease although a wide range of enzymatic and clinical variability occurs which may be due to tissue-specific alternative splicing. Alternatively spliced transcripts encoding different isoforms have been described. [provided by RefSeq]
