Western Blot: alpha Tubulin Antibody (F2C) - Chimeric [NBP3-09006] - HeLa (A), HEK293 (B), and HepG2 (C) cell lysate samples (35ug protein in RIPA buffer) were resolved on a 10% SDS PAGE gel and blots probed with the chimeric rabbit version of F2C (NBP3-09006) at 0.01 ug/ml before detection using an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence. The expected band size for alpha Tubulin is ~55kDa. NBP3-09006 successfully detected human alpha Tubulin in HeLa, HEK293, and HepG2 cell lysate samples.

Immunohistochemistry-Paraffin: alpha Tubulin Antibody (F2C) - Chimeric [NBP3-09006] - Anti-Alpha Tubulin staining of paraffin embedded human cerebral cortex tissue using the rabbit-chimeric version of F2C (NBP3-09006). Antigen retreival was acheived by microwaving in citrate buffer (pH6), followed by blocking with protein block serum-free buffer. Primary antibody incubation with NBP3-09006 was carried out at 4 ug/ml for 30 minutes. Samples were then incubated with an anti-rabbit IgG HRP secondary antibody for 20 mins followed by DAB (3,3'-diaminobenzidine), and counter-staining with haemotoxylin. Staining of axons and a few neuronal cell bodies may be observed. Recommended concentration, 2-4 ug/ml.

This antibody binds alpha-tubulin. Tubulin is the major constituent of microtubules. The alpha-chain has a non-exchangeable GTP-binding site.

Background: alpha Tubulin

Tubulin is the major building block of microtubules. Microtubules function as structural and mobile elements in mitosis, intracellular transport, flagellar movement and in the cytoskeleton. The tubulin superfamily is composed of six distinct families. Genes from the alpha, beta and gamma tubulin families are found in all eukaryotes. Microtubules of the eukaryotic cytoskeleton perform essential and diverse functions and are composed of a heterodimer of alpha and beta tubulins. Alpha and beta tubulin dimers are assembled to 13 protofilaments that form a microtubule of 22 nm diameter with a theoretical molecular weight of 50 kDa.

Tyrosine ligase adds a C-terminal tyrosine to monomeric alpha tubulin. Assembled microtubules can again be detyrosinated by a cytoskeleton associated carboxypeptidase. Detyrosinated alpha tubulin is referred to as Glu-tubulin. Another post-translational modification of detyrosinated alpha tubulin is C-terminal polyglutamylation which is characteristic for microtubules in neuronal cells and the mitotic spindle.

Like GAPDH and beta-actin, alpha/beta tubulin is often used as a loading control in immunoblot applications (1). Alpha/beta tubulin is also good for counterstaining microtubules in immunofluorescence (2).
