Immunocytochemistry/Immunofluorescence: ATP7b Antibody (9H8) [NBP3-26489] - Staining of Hela cell with ATP7b Antibody (9H8) at 1:100, counter-stained with DAPI. The cells were fixed in 4% formaldehyde and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4C. The secondary antibody was Alexa Fluor 488-conjugated Goat Anti-Rabbit IgG (H+L).

Then 10% normal goat serum was Incubated to block non-specific protein-protein interactions followed by the antibody (1ug/1*10^6 cells) for 45 min at 4C. The secondary antibody used was FITC-conjugated Goat Anti-Rabbit IgG (H+L) at 1/200 dilution for 35 min at 4C. Isotype control antibody (green line) was mouse IgG1 (1ug/1*10^6 cells) used under the same conditions. Acquisition of >10,000 events was performed.

Background: ATP7b

ATP7b is an important protein for copper transport and elimination of excess copper from the body. ATP7b transports metals in and out of cells using ATP. There are 3 known isoforms of the ATP7b gene; form A is found in the liver, kidney, and brain, the shorter form B is found in brain tissue, and the third isoform, known as WND/140 KDA is found in mitochondria. Mutations in the ATP7b gene can cause Wilson's disease, an inherited disorder causing copper poisoning in the brain and liver.
