Clone (M016) was generated from a proprietary antigen related to the native human ATPase Na+/K+ beta 3 subunit expressed in A431 epidermoid carcinoma cell line. [UniProt# P54709]

Specificity

Clone M016 mouse monoclonal antibody detects a 40 kDa protein on SDS-PAGE "Native" immunoblots of human A431, LNCaP, MeWo, MDA-MB-231, and MCF7 cells. This antibody does not detect denatured ATPase Na+/K+ beta 3 subunit. In addition, mass spectrometry analysis of immunoprecipitates using NM0161 in human A431 cell lysates confirms that this antibody only detects ATPase Na+/K+ beta 3 protein.

Immunocytochemistry/Immunofluorescence: ATPase Na+/K+ beta 3 Antibody (M016) [NBP3-23433] - Immunocytochemical labeling of ATPase Na+/K+ beta 3 in paraformaldehyde fixed human MDA-MB-231 cells. The cells were labeled with mouse monoclonal anti-ATPase Na+/K+ beta 3 (clone M016). The antibody was detected using goat anti-mouse Ig DyLight(R) 594.

The NBP3-23433 antibody only (lane 1), IP from A431 cell lysate (lane 2), A431 cell input (lane 3), LNCaP cells (lane 4), MeWo cells (lane 5), and normal human lung (lane 6) and skin (lane 7). The blot was probed with anti-ATPase Na+/K+ beta 3 NBP3-23433 (lanes 1-7). The arrow designates native antibody, while the beta 3 subunit migrates around 40 kDa.

Background: ATPase Na+/K+ beta 3

ATPase Na+/ K+ beta 3 is encoded by this gene belongs to the family of Na+/K+ and H+/K+ ATPases beta chain proteins, and to the subfamily of Na+/K+ -ATPases. Na+/K+ -ATPase is an integral membrane protein responsible for establishing and maintaining the electrochemical gradients of Na and K ions across the plasma membrane. These gradients are essential for osmoregulation, for sodium-coupled transport of a variety of organic and inorganic molecules, and for electrical excitability of nerve and muscle. This enzyme is composed of two subunits, a large catalytic subunit (alpha) and a smaller glycoprotein subunit (beta). The beta subunit regulates, through assembly of alpha/beta heterodimers, the number of sodium pumps transported to the plasma membrane. The glycoprotein subunit of Na+/K+ -ATPase is encoded by multiple genes. This gene encodes a beta 3 subunit. This gene encodes a beta 3 subunit. A pseudogene exists for this gene, and it is located on chromosome 2. [provided by RefSeq]
