Phospho-Actin (Tyr-53) synthetic peptide (coupled to KLH) corresponding to amino acid residues around tyrosine 53 of human beta-Actin. This sequence is identical to similar regions in all four alpha-actins, as well as in gamma-actin, and is well conserved in actins from most eukaryotic species. [UniProt# P60709]

Specificity

This antibody detects a 42 kDa protein corresponding to the molecular mass of Actin on SDS-PAGE immunoblots of pervanadate treated human C2C12 and SYF cSrc transformed cells, but not in control cells. In addition, this antibody either detects actin cleavage products at 36 and 20 kDa, or cross-reacts with unidentified proteins at these molecular weights in C2C12 cells.

Immunocytochemistry/Immunofluorescence: beta-Actin [p Tyr53] Antibody [NBP3-23323] - Immunocytochemical labeling using anti-Actin (N-terminal) and anti-Actin (Tyr-53) polyclonal antibodies in C2C12 cells control (left) or treated with pervanadate (1 mM) for 30 min (middle). The cells were fixed in paraformaldehyde and permeabilized in acetone. Both antibodies were used in the presence of blocking peptide: Actin (N-terminal) peptide or phospho-Actin (Tyr-53) peptide, respectively (right).

Background: beta-Actin

Actin is a highly conserved protein involved in cell motility, structure, and integrity. Six functional actin isoforms exist and were named based on their electrophoretic mobility and distribution including alpha-cardiac muscle actin (aCAA), alpha-skeletal muscle actin (aSKA), alpha-smooth muscle actin (aSMA), beta-cytoplasmic actin (bCYA), gamma-cytoplasmic actin (gCYA) and gamma-smooth muscle actin (gSMA) (1). The cytoplasmic isoforms, beta-actin and gamma-actin, are nearly identical, differing by only 4 amino acids at the N-terminus with a theoretical molecular weight of 42 kDa (2). Beta-actin is ubiquitously expressed in eukaryotic cells and considered a housekeeping gene which is frequently used as a loading control for assays involving protein detection, such as Western blotting (3,4).
