BIN1 Antibody (99D) was produced in mouse by repeated immunizations with a fragment portion of recombinant human BIN1 protein followed by hybridoma development. (Uniprot: O00499)

Reactivity Notes

A BLAST analysis was used to suggest cross-reactivity with BIN1 from human and mouse sources based on 100% homology with the immunizing sequence. Cross-reactivity with BIN1 from other sources has not been determined.

This antibody was purified from clarified mouse ascetic fluid by Protein A chromatography followed by extensive dialysis against the buffer stated above

Store vial at -20C prior to opening. Aliquot contents and freeze at -20C or below for extended storage. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4C as an undiluted liquid. Dilute only prior to immediate use.

Background: BIN1

BIN1 encodes several isoforms of a nucleocytoplasmic adaptor protein, one of which was initially identified as a MYC-interacting protein with features of a tumor suppressor. Isoforms that are expressed in the central nervous system may be involved in synaptic vesicle endocytosis and may interact with dynanim, synaptojanin, endophilin, and clathrin. Isoforms that are expressed in muscle and ubiquitously expressed isoforms localize to the cytoplasm and nucleus and activate a caspase independent apoptotic process. Studies in mouse suggest that this gene plays an important role in cardiac muscle development. Alternate splicing of the gene results in ten transcript variants encoding different isoforms. Aberrant splice variants expressed in tumor cell lines have also been described.
