Flow Cytometry: BubR1 Antibody (8G1) - Azide and BSA Free [NBP2-80606] - An intracellular stain was performed on HeLa cells with BubR1 Antibody (8G1)NB100-353C (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 5 ug/mL for 30 minutes at room temperature. Both antibodies were directly conjugated to Dylight 650.

Background: BubR1

BubR1 (budding uninhibited by benzimidazole-related 1 or mitotic checkpoint serine/threonine-protein kinase BUB1 beta) is an essential component of mitotic checkpoint/control complex which is required for normal mitotic progression and any defects in this complex may result in chromosomal instability/aberrant mitosis. This evolutionarily conserved and crucial system of self-control in cell cycle warrants high quality of chromosomal segregation by postponing manifestation of anaphase in all chromosomes which are incorrectly bi-oriented to mitotic spindles. One of its checkpoint functions is to prevent the activity of anaphase-promoting complex/cyclosome (APC/C) by blocking the binding of CDC20 to APC/C, independently of its kinase activity; and the other is to monitor kinetochore activities that depend on the kinetochore motor CENPE. BubR1 is required for kinetochore localization of CENPE and it negatively regulates PLK1 activity in interphase cells and suppresses centrosome amplification. BubR1 is also implicated in triggering apoptosis in polyploid cells that exit aberrantly from mitotic arrest and thus it behaves as a tumor suppressior. Defects in BubR1 have been associated with tumor formation and are the underlying cause of premature chromatid separation trait (PCS) as well as mosaic variegated aneuploidy syndrome type 1 (MVA1).
