Immunohistochemistry: Calcitonin Antibody (PD00-46) [NBP3-32101] - Immunohistochemical analysis of paraffin-embedded human medullary thyroid carcinoma tissue with Rabbit anti-Calcitonin antibody (NBP3-32101) at 1/20,000 dilution.

The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (NBP3-32101) at 1/20,000 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Background: Calcitonin

Calcitonin is a 32 amino acid peptide hormone synthesized by the parafollicular cells of the thyroid. It causes a rapid, but short lived, reduction in serum calcium and phosphate by promoting the incorporation of those ions in the bones. This effect is opposite to that of parathyroid hormone. Staining for calcitonin may be used for the identification of a spectrum of C cell proliferative abnormalities ranging from C cell hyperplasia to invasive tumors. Staining for calcitonin in medullary carcinoma of the thyroid produces a fine granular pattern in the cytoplasm. Amyloid deposits within the tumor may also exhibit varying degrees of calcitonin activity.
