Chromatin from K562 cells was immunoprecipitated with anti-CBX2 antibody NBP1-40342 and analyzed by DNA sequencing. The figure illustrates the peak distribution of CBX2 binding within a 1.5 Mb region of chromosome 1 as detected using anti-CBX2 antibody NBP1-40342. Immunoprecipitation: Cbx2 Antibody [NBP1-40342] - Detection of human CBX2 by western blot of immunoprecipitates. Samples: Whole cell lysate (0.5 or 1.0 mg per IP reaction; 20% of IP loaded) from HEK293T cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-CBX2 antibody NBP1-40342 (lot NBP1-40342-3) used for IP at 6 ug per reaction. CBX2 was also immunoprecipitated by previous lots of this antibody (lots NBP1-40342-1 and NBP1-40342-2). For blotting immunoprecipitated CBX2, NBP1-40342 was used at 1 ug/ml. Detection: Chemiluminescence with an exposure time of 30 seconds.

Background: Cbx2

Polycomb' genes in Drosophila maintain the repressed state of homeotic and other developmentally regulatedgenes (Wedeen et al., 1986 (PubMed 3081265); Kuziora and McGinnis, 1988 (PubMed 2903050)) by mediating changes inhigher-order chromatin structure (Paro and Hogness, 1991 (PubMed 1898775)). M33, a mouse homolog of polycomb, wasisolated by means of the structural similarity of its chromodomain (Pearce et al., 1992 (PubMed 1352241)). The fifthexon of M33 contains a region of homology shared by Drosophila and Xenopus. In Drosophila, its deletion results in theloss of polycomb function.(supplied by OMIM)
