This product is an affinity purified antibody produced by immunoaffinity chromatography using phospho peptide coupled to agarose beads followed by solid phase adsorption(s) against non-phospho peptide and non-specific peptide to remove any unwanted reactivities. This antibody is specific for phosphorylated human CDC16 protein at the pT580 residue. BLAST analysis indicates 100 % homology of the immunizing sequence with CDC16 from human. No cross reactivity is expected against homologues from other sources. This antibody is expected to react with all isoforms of this protein. Minimal reactivity is expected with the non-phosphorylated form of the protein.

Store vial at -20C prior to opening. Aliquot contents and freeze at -20C or below for extended storage. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4C as an undiluted liquid. Dilute only prior to immediate use.

Western Blot: CDC16 [p Thr580] Antibody [NB600-889] - Analysis of a band 72 kDa corresponding to phosphorylated human APC6 (arrowhead lane 1). Lane 1 - nocodazole treated HeLa whole cell lysate. Lane 2 - Reactivity is not seen in lysates from asynchronous HeLa whole cell cultures. Each lane contains approximately 35ug of lysates, separated by 4-20% SDS-PAGE Tris-HEPES and then transferred to nitrocellulose. After blocking the membrane was probed with the primary antibody diluted to 1:1,000 overnight at 4C followed by washes and reaction with a 1:10,000 dilution of IRDye800 conjugated Gt-a-Rabbit IgG [H&L] MX for 45 min at room temperature.

Background: CDC16

APC6 (anaphase-promoting complex subunit 6) is a member of the E3 enzyme family. This protein contains TPR repeats and has a molecular weight of approximately 72 kD. The APC6 protein is located at the centrosome and mitotic spindle. This protein is required for core stability in the spindle checkpoint pathway. The APC6 protein functions with other members of the APC complex as a multisubunit cell cycle ubiquitin ligase, and a regulator of sister chromatid separation by degrading securins. In addition, this protein functions in ubiquitin-dependent cyclin catabolism, metaphase/anaphase transition and spindle elongation. The APC6 protein comprises one subunit of the anaphase promoting complex including APC1-8 and other probable complex proteins APC9-11, Cdc26, Mnd2, Swm1. The APC complex is inactivated by protein kinase A and is activated by CDC20 and Cdh1.
