Immunoprecipitation: CDCA3 Antibody [NB100-74621] - Detection of Human CDCA3/TOME1 by Western Blot of Immunoprecipitates. Samples: Whole cell lysate (1 mg for IP, 20% of IP loaded) from HeLa cells. Antibodies: Affinity purified rabbit anti-CDCA3/TOME1 antibody NB100-74621 used for IP at 3 mcg/mg lysate. CDCA3/TOME1 was also immunoprecipitated by rabbit anti-CDCA3/TOME1 antibody NB100-74622, which recognizes a downstream epitope. Detection: Chemiluminescence with an exposure time of 3 minutes.

Background: CDCA3

CDCA3/TOME1 is an F-box protein and is part of the SCF complex, an ubiquitin ligase complex that promotes degradation of the mitosis-inhibitory kinase, wee1. CDCA3/TOME1 expression is cell cycle regulated by cis-promoter elements and activated at the G2/M phase. CDCA3/TOME1 levels are also regulated at the protein level via degradation by the APC (anaphase-promoting complex) during G1.
