Background: ch-TOG

Ch-TOG (colonic and hepatic tumor overexpressed gene protein), also called cytoskeleton-associated protein 5 (CKAP5) is a member of TOG/XMAP215 family which associates with the plus end of microtubules and regulates microtubule dynamics/organization. Ch-TOG promotes cytoplasmic microtubule nucleation and elongation, and plays a major role in organizing spindle poles. The N-terminal half of Ch-TOG contains a microtubule-binding domain whereas the C-terminal half contains a KXGS motif for binding the tubulin dimers. In spindle formation, Ch-TOG protects kinetochore microtubules from depolymerization and plays an essential role in centrosomal microtubule assembly. Moreover, Ch-TOG is essential to proper interaction of microtubules with the cell cortex for directional cell movement. Ch-TOG also implicates in translation of MBP-mRNA by interacting with heterogeneous nuclear ribonucleoprotein (hnRNP) A2. Ch-TOG interacts with SLAIN2 also and can be detected on centrosomes during interphase or on the spindle poles as well as microtubules during mitosis. Ch-TOG is expressed in several normal tissues including skeletal muscle, brain, heart, placenta, lung, liver, kidney, pancreas etc. and is found overexpressed in hepatomas and colonic cancers.
