Application Notes
This ChABC (1E10) antibody is useful for ELISA and Western blot, where a band can be seen at approximately 80 kDa. It can be used in Sandwich ELISA as the detection antibody with catalog # NBP1-96142, ChABC (6A12) as the capture antibody. The observed molecular weight of the protein may vary from the listed predicted molecular weight due to post translational modifications, post translation cleavages, relative charges, and other experimental factors. Western blot data: 125 ng Chondroitinase ABC was mixed with 100 ug of fetal bovine serum (as a background control). The proteins were mixed with 100 ul of non-reducing sample buffer and applied to a single 6 cm well set in a SDS-PAGE minigel (approx 20 ng/well [0.5 cm]). The gel bands were electroblotted to a nitrocellulose sheet. Immunolabeling for individual antibodies was performed on the sheet placed in a slot blot devise. Purified NBP1-96141 (ChABC/1E10) and NBP1-96142 (ChABC/6A12) were applied at 5 ug/ml IgG.

Background: ChABC

ChABC (Chondroitinase ABC) is a bacterial enzyme that has been used for decades, mostly for biochemical characterization of chondroitin sulfate proteoglycans (CSPG). This enzyme degrades the sugar chains attached to the proteoglycan core protein. More recently, ChABC has been used to therapeutically degrade and inactivate CSPG in vivo. CSPGs are involved in a variety of biological processes and play a major inhibitory role in regeneration in the nervous system. For instance, CSPGs have been implicated in preventing regeneration after spinal cord injury. Injection of ChABC at the site of spinal cord injury has improved recovery in numerous animal models. ChABC antibodies serve as tools to help determine the ChABC distribution and content in tissues.
