Specificity

This monoclonal antibody reacts with the rod domain of human cytokeratin-19 (CK19), a polypeptide of 40kDa. Its epitope maps between amino acid 312-335. CK19 is expressed in sweat gland, mammary gland ductal and secretory cells, bile ducts, gastrointestinal tract, bladder urothelium, oral epithelia, esophagus, and ectocervical epithelium. Anti-CK19 reacts with a wide variety of epithelial malignancies including adenocarcinomas of the colon, stomach, pancreas, biliary tract, liver, and breast. Perhaps the most useful application is the identification of thyroid carcinoma of the papillary type, although 50%-60% of follicular carcinomas are also labeled. Anti-CK19 is a useful marker for detection of tumor cells in lymph nodes, peripheral blood, bone marrow and breast cancer.

Application Notes
Immunohistochemistry (Formalin-fixed): 1-2ug/ml for 30 minutes at RT. Staining of formalin-fixed tissues requires heating tissue sections in 10mM Tris with 1mM EDTA, pH 9.0, for 45 min at 95C followed by cooling at RT for 20 minutes.
Optimal dilution for a specific application should be determined.
Please Note: Optimal dilutions of this antibody should be experimentally determined.

Background: Cytokeratin 19

Keratin-19 (K19) is a type I intermediate filament protein and one of the best characterized of the keratins expressed in mature striated muscle. Keratin-19 is expressed in a defined zone of basal keratinocytes in the deep outer root sheath of hair follicles. Also observed in sweat gland and mammary gland ductal and secretory cells, bile ducts, gastrointestinal tract, bladder urothelium, oral epithelia, esophagus, ectocervical epithelium. Keratin-19 has been used as a useful marker for detection of tumor cells in lymph nodes, peripheral blood, bone marrow and breast cancer. Immunohistochemical data has shown that Keratin-19 may be uses as a marker for human skin stem cells. This antibody reacts with the rod domain of human Keratin-19, a polypeptide of 40kDa. The antibody recognition epitope maps between amino acid 312-335. This antibody reacts with the MCF-7 cells which are known to contain Keratin-19. It shows no reaction with the cells lacking Keratin-19 such as A431 and HeLa. Keratin-19 is not expressed in hepatocytes, therefore antibody to Keratin-19 is useful in the identification of liver metastasis.
