Background: DUOX2

DUOX2 (dual oxidase 2), a multi-pass membrane protein localized to the apical membrane of epithelial cells, belongs to NADPH oxidase (NOX) family which is known to reduce molecular oxygen with NADPH as electron donor and unlike most of NOX-family members that produce superoxide anions (O2.-) as primary product, DUOX directly produce H2O2. DUOX2 and DUOX 1 require specific maturation factors, DUOXA1 and DUOXA2, to be functionally expressed at the apical follicular membrane, the site for hormonogenesis. DUOX2/DUOX 1 are major components of H2O2-generating system and generate H2O2 which is required for activity of thyroid peroxidase/TPO and lactoperoxidase/LPO. DUOX2 plays a role in thyroid hormones synthesis and LPO-mediated antimicrobial defense mucosal surface. It has its own peroxidase activity through its N-terminal peroxidase-like domain which is inhibited by aminobenzohydrazide and the NADPH oxidase activity is calcium-dependent. DUOX2 is expressed in thyrocytes, intestine, kidney, liver, lung, pancreas, prostate, salivary glands, testis etc. and is induced by forskolin, thyrotropin and the Th1-specific cytokine IFNG/IFN-gamma. Defects in DUOX2 have been linked to thyroid dyshormonogenesis 6 (TDH6) characterized by defective conversion of accumulated iodide to organically bound iodine.
