Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (NBP3-32283) at 1/1,000 dilution was used in 5% NFDM/TBST at 4C overnight. Goat Anti-Mouse IgG - HRP Secondary Antibody at 1/50,000 dilution was used for 1 hour at room temperature.

Immunohistochemistry: Dynamin Antibody (A1B2-R) [NBP3-32283] - Immunohistochemical analysis of paraffin-embedded human brain tissue with Mouse anti-Dynamin antibody (NBP3-32283) at 1/1,000 dilution.

The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (NBP3-32283) at 1/1,000 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Background: Dynamin

The dynamin family of GTPases is essential for receptor-mediated endocytosis and synaptic vesicle recycling, and it has recently been shown to play a role in vesicle formation from the trans-Golgi network (1). Dynamin-1 is a GTP-, microtubule-, and phospholipid-binding protein that is expressed primarily in brain and almost exclusively expressed in the central nervous system (2). Dynamin-1 is believed to assemble around the necks of clathrin-coated pits and assist in pinching vesicles from the plasma membrane. This role would make dynamin unique among GTPases in its ability to act as a mechanochemical enzyme (1). Dynamin-1 function is apparently essential for scission of newly formed vesicles from the plasma membrane; it has been proposed that dynamin is a regulator of downstream effectors of scission, rather than being directly responsible for it (3).
