Immunohistochemistry: ER alpha/NR3A1 Antibody (PD00-04) [NBP3-32321] - Immunohistochemical analysis of paraffin-embedded human cervix tissue with Rabbit anti-ER alpha/NR3A1 antibody (NBP3-32321) at 1/1,000 dilution.

The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (NBP3-32321) at 1/1,000 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Background: ER alpha/NR3A1

Estrogen receptor alpha (ER alpha) is a nuclear protein and member of the steroid hormone receptor family. ER alpha possesses both DNA binding and ligand binding domains, and exerts a significant role in activating the transcription of certain genes (1). Ligand-dependent dimerization and phosphorylation on serines, 104, 106, and 118 function to regulate the transcriptional activation of ER alpha (2). The phosphorylation of the human estrogen receptor (ER) serine residue at position 118 is required for full activity of the ER activation function 1 (AF-1) (3).
