Antibody: Affinity purified rabbit anti-G9A/EHMT2 antibody NB100-40825 (lot NB100-40825-2) used for IP at 6 ug per reaction. G9A/EHMT2 was also immunoprecipitated by a previous lot of this antibody (NB100-40825-1) and another rabbit anti-G9A/EHMT2 antibody. For blotting immunoprecipitated G9A/EHMT2, NB100-40825 was used at 1 ug/ml. Detection: Chemiluminescence with an exposure time of 3 minutes.

While trace amounts of G9A/EHMT2 can be immunoprecipitated by this antibody, the efficiency is very low. Epitope retrieval with Tris-EDTA pH9.0 is recommended for FFPE tissue sections.

Background: G9a/EHMT2

G9A antibody contains ankyrin repeats, suggesting involvement in intracellular protein-protein interaction. G9A is a histone methyltransferase that preferentially methylates Lys-9 of histone H3 and Lys-27 of histone H3 (in vitro). H3 Lys-9 methylation represents a specific tag for epigenetic transcriptional repression by recruiting HP1 proteins to methylated histones. G9A is probably targeted to histone H3 by different DNA-binding proteins like E2F6, MGA, MAX and/or DP1.
