Background: Gephyrin/GPHN

The Gephyrin (GPHN) gene encodes a neuronal assembly protein that exists in isoform 1 (736 amino acids long, 79 kDA) or isoform 2 (769 amino acids in length, approximately 83 kDA). This protein anchors inhibitory neurotransmitter receptors to the postsynaptic cytoskeleton through high affinity binding to a receptor subunit domain and tublin dimers. The protein coded by GPHN is also necessary for molybdenum biosynthesis. The GPHN gene has been investigated in relation to many diseases such as leukemia, cervicitis, tuberculosis, alzheimer's disease, anorexia nervosa, huntington's disease, retinitis, and herpes zoster. The GPHN gene interacts with genes DYNLL1, PFN1, PRPF4, ENAH, and SLX4 to participate in pathways that regulate metabolism of vitamins and cofactors as well as molybdenum cofactor biosynthesis.
