Immunoprecipitation:GGA1 AntibodyNBP3-30196] -Detection of human GGA1 by western blot of immunoprecipitates. Samples: Whole cell lysate (0.5 or 1.0 mg per IP reaction; 20% of IP loaded) from 293T cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-GGA1 antibody NBP3-30196 (lot NBP3-30196-1) used for IP at 6 µg per reaction. GGA1 was also immunoprecipitated by rabbit anti-GGA1.. For blotting immunoprecipitated GGA1, NBP3-30196 was used at 0.4 µg/ml. Detection: Chemiluminescence with an exposure time of 10 seconds.

Background: GGA1

GGA1 encodes a member of the Golgi-localized, gamma adaptin ear-containing, ARF-binding (GGA) protein family. Members of this family are ubiquitous coat proteins that regulate the trafficking of proteins between the trans-Golgi network and the lysosome. These proteins share an amino-terminal VHS domain which mediates sorting of the mannose 6-phosphate receptors at the trans-Golgi network. They also contain a carboxy-terminal region with homology to the ear domain of gamma-adaptins. Multiple alternatively spliced transcript variants encoding different isoforms have been found for this gene.
