Immunocytochemistry/Immunofluorescence: GITR/TNFRSF18 Antibody (YGITR765) [NBP2-81090] - Immunofluorescence staining of fixed mouse splenocytes with GITR/TNFRSF18 Antibody (YGITR765) [NBP2-81090]. Immunofluorescence analysis of paraformaldehyde fixed mouse (Mus musculus) splenocytes on Shi-fix coverslips, permeabilized with 0.15% Triton stained with the chimeric rabbit version of YGITR765 [NBP2-81090] at 10 ug/ml for 1h followed by Alexa Fluor 488 secondary antibody (1 ug/ml), showing membrane staining in a subset of cells. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom NBP2-81090, DAPI, merged channels and an isotype control. The isotype control was stained with an anti-Fluorescein antibody followed by Alexa Fluor 488 secondary antibody.

Flow Cytometry: GITR/TNFRSF18 Antibody (YGITR765) [NBP2-81090] - Flow-cytometry using [NBP2-81090]. Mouse activated T cells were stained with an isotype control (NBP2-52638, Orange) or the rabbit-chimeric version of YGITR765 [NBP2-81090] at a concentration of 1 ug/ml for 30 mins at RT without (Blue) and with (Grey) stimulation of the lymphocytes with immobilized anti-CD3 and anti-CD28 antibodies. After washing, bound antibody was detected using a AF488 conjugated donkey anti-rabbit antibody and cells analysed on a FACSCanto flow-cytometer.

Application Notes
This chimeric rabbit antibody was made using the variable domain sequences of the original Rat IgG2b format, for improved compatibility with existing reagents, assays and techniques. A chimeric version of this antibody featuring a human IgG1 Fc-portion engineered to lack the N-linked glycosylation site at amino acid 297 was shown to enhance humoral immune responses by blocking GITR signallin (Ponte 2010).

Background: GITR/TNFRSF18

GITR is encoded by this gene is a member of the TNF-receptor superfamily. This receptor has been shown to have increased expression upon T-cell activation, and it is thought to play a key role in dominant immunological self-tolerance maintained by CD25(+)CD4(+) regulatory T cells. Knockout studies in mice also suggest the role of this receptor is in the regulation of CD3-driven T-cell activation and programmed cell death. Three alternatively spliced transcript variants of this gene encoding distinct isoforms have been reported. [provided by RefSeq]
