Western Blot: Glutamate Receptor 4 Antibody (JE36-81) [NBP3-32384] - Western blot analysis of Glutamate Receptor 4 on different lysates with Rabbit anti-Glutamate Receptor 4 antibody (NBP3-32384) at 1/1,000 dilution.

Lane 1: Mouse cerebellum tissue lysate (RIPA lysis)
Lane 2: Mouse cerebellum tissue lysate (hot lysis)
Lane 3: Rat cerebellum tissue lysate (RIPA lysis)
Lane 4: Rat cerebellum tissue lysate (hot lysis)

4-20% SDS-PAGE gel.

Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (NBP3-32384) at 1/1,000 dilution was used in 5% NFDM/TBST at 4 overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1:100,000 dilution was used for 1 hour at room temperature.

Flow Cytometry: Glutamate Receptor 4 Antibody (JE36-81) [NBP3-32384] - Flow cytometric analysis of SH-SY5Y cells labeling Glutamate Receptor 4.

Cells were fixed and permeabilized. Then stained with the primary antibody (NBP3-32384, 1ug/ml) (red) compared with Rabbit IgG Isotype Control (green). After incubation of the primary antibody at +4℃ for an hour, the cells were stained with a iFluor™ 488 conjugate-Goat anti-Rabbit IgG Secondary antibody at 1/1,000 dilution for 30 minutes at +4℃. Unlabelled sample was used as a control (cells without incubation with primary antibody; black).

Background: GluR4

Glutamate receptors mediate most excitatory neurotransmission in the brain and play an important role in neural plasticity, neural development and neurodegeneration. Ionotropic glutamate receptors are categorized into NMDA receptors and kainate/AMPA receptors, both of which contain glutamategated, caution-specific ion channels. Kainate/AMPA receptors are co-localized with NMDA receptors in many synapses and consist of seven structurally related subunits designated GluR-1 to -7. The kainate/AMPA receptors are primarily responsible for the fast excitatory neuro-transmission by glutamate, whereas the NMDA receptors are functionally characterized by a slow kinetic and a high permeability for Ca2+ ions. The NMDA receptors consist of five subunits: epsilion 1, 2, 3, 4 and one zeta subunit. The zeta subunit is expressed throughout the brain stem, whereas the four epsilon subunits display limited distribution.
