Lane 3: Rat brain tissue lysate (10 ug/Lane)

Predicted band size: 103 kDa
Observed band size: 103 kDa

Exposure time: 53 seconds;

4-20% SDS-PAGE gel.

Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (NBP3-32405) at 1/5,000 dilution was used in 5% NFDM/TBST at 4 overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1:50,000 dilution was used for 1 hour at room temperature.
Glutamate Receptor 6 Antibody (JE39-33)

Immunohistochemistry: Glutamate Receptor 6 Antibody (JE39-33) [NBP3-32405]

Immunohistochemistry: Glutamate Receptor 6 Antibody (JE39-33) [NBP3-32405] - Immunohistochemical analysis of paraffin-embedded human brain tissue with Rabbit anti-Glutamate Receptor 6 antibody (NBP3-32405) at 1/1,000 dilution.

The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (NBP3-32405) at 1/1,000 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Background: GluR6/GRIK2

Glutamate receptors mediate most excitatory neurotransmission in the brain and play an important role in neural plasticity, neural development and neurodegeneration. Ionotropic glutamate receptors are categorized into NMDA receptors and kainate/AMPA receptors, both of which contain glutamategated, caution-specific ion channels. Kainate/AMPA receptors are co-localized with NMDA receptors in many synapses and consist of seven structurally related subunits designated GluR-1 to -7. The kainate/AMPA receptors are primarily responsible for the fast excitatory neuro-transmission by glutamate, whereas the NMDA receptors are functionally characterized by a slow kinetic and a high permeability for Ca2+ ions. The NMDA receptors consist of five subunits: epsilion 1, 2, 3, 4 and one zeta subunit. The zeta subunit is expressed throughout the brain stem, whereas the four epsilon subunits display limited distribution.
