Description

This antibody is an IgG fraction antibody purified from monospecific antiserum by a multi-step process which includes delipidation, salt fractionation and ion exchange chromatography followed by extensive dialysis against the buffer stated above. Assay by immunoelectrophoresis resulted in a single precipitin arc against anti-Goat Serum as well as purified and partially purified Glycerol Kinase [Cellulomonas species]

Store vial at 4C prior to restoration. For extended storage aliquot contents and freeze at -20C or below. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4C as an undiluted liquid. Dilute only prior to immediate use.

Western Blot: Glycerol Kinase Antibody [NBP1-78018] - Glycerol Kinase antibody was used to detect purified Glycerol Kinase under reducing (R) and non-reducing (NR) conditions. Reduced samples of purified protein contained 4% BME and were boiled for 5 minutes. Samples of ~1ug of protein per lane were run by SDS-PAGE. Protein was transferred to nitrocellulose and probed with 1:3000 dilution of primary antibody (block 4C). Detection shown was using Dylight 488 conjugated Donkey anti goat (1:10K in TBS/blocking buffer 1 hr RT). Images were collected using the VersaDoc System.

Primary antibody: Glycerol Kinase Antibody Biotin Conjugated at 1:1000 overnight at 4C. Secondary antibody: HRP Streptavidin secondary antibody at 1:40,000 for 30 min at RT. Block: incubated with blocking buffer for 30 min at RT. Predicted/Observed size: 55 kDa, 55 kDa for Glycerol Kinase.

Western Blot: Glycerol Kinase Antibody [NBP1-78018] - Both the antiserum and IgG fractions of Glycerol Kinase antibody) are shown to detect the 128,000 dalton enzyme in cellular extracts. Approximately 10 ug of total protein is loaded per lane. A 1:4,000 dilution of the primary antibody is used followed by detection using HRP Rabbit-a-Goat IgG [H&L] diluted 1:4,000 and color development using 4-CN substrate until sufficient color develops. Other detection systems will yield similar results.

Application Notes
This product has been tested by ELISA and western blot. This product is assayed against 1.0 ug of Glycerol Kinase [Cellulomonas species] in a standard ELISA using Peroxidase conjugated Affinity Purified anti-Goat IgG [H&L] (Rabbit) and (ABTS (2,2'-azino-bis-[3-ethylbenthiazoline-6-sulfonic acid]) as a substrate for 30 minutes at room temperature. A working dilution of 1:4,000 to 1:18,000 of the reconstitution concentration is suggested for this product.

Background: Glycerol Kinase

Glycerol kinase catalyzes the formation of glycerol 3 phosphate from ATP and glycerol. Dihydroxyacetone and L glyceraldehyde can also act as acceptors; UTP and, in the case of the yeast enzyme, ITP and GTP can act as donors. It provides a way for glycerol derived from fats or glycerides to enter the glycolytic pathway. NB - Glycerol kinase obtained from Cellulomonas spp has an observed molecular weight of 128kDa.
