Immunoprecipitation:HADHA AntibodyNBP3-28907] -Detection of human HADHA by western blot of immunoprecipitates. Samples: Whole cell lysate (0.5 or 1.0 mg per IP reaction; 20% of IP loaded) from 293T cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-HADHA antibody NBP3-28907 (lot NBP3-28907-1) used for IP at 6 µg per reaction. HADHA was also immunoprecipitated by rabbit anti-HADHA antibody. For blotting immunoprecipitated HADHA, NBP3-28907 was used at 0.4 µg/ml. Detection: Chemiluminescence with an exposure time of 10 seconds.

Background: HADHA

HADHA encodes the alpha subunit of the mitochondrial trifunctional protein, which catalyzes the last three steps of mitochondrial beta-oxidation of long chain fatty acids. The mitochondrial membrane-bound heterocomplex is composed of four alpha and four beta subunits, with the alpha subunit catalyzing the 3-hydroxyacyl-CoA dehydrogenase and enoyl-CoA hydratase activities. Mutations in this gene result in trifunctional protein deficiency or LCHAD deficiency. The genes of the alpha and beta subunits of the mitochondrial trifunctional protein are located adjacent to each other in the human genome in a head-to-head orientation.
