Immunocytochemistry/Immunofluorescence: hHR23b Antibody (SAIC-28B-8) - Azide and BSA Free [NBP3-20097] - Immunofluorescence analysis of paraformaldehyde fixed HeLa cells permeabilized with 0.15% Triton stained with the chimeric rabbit IgG version of SAIC-28B-8 (NBP3-20097) (1:100 dilution) for 1h followed by Alexa Fluor(R) 488 secondary antibody (1:1500 dilution), showing nuclear and cytoplasmic staining. Actin filaments were stained with phalloidin (red) and the nuclear stain is DAPI (blue). Panels show from left-right, top-bottom NBP3-20097, DAPI, merged channels and an isotype control. The isotype control was an unknown specificity antibody followed by staining with Alexa Fluor(R) 488 secondary antibody.

Western Blot: hHR23b Antibody (SAIC-28B-8) - Azide and BSA Free [NBP3-20097] - Nuclear lysate of MCF7 cells (35ug protein in RIPA buffer) were resolved on a SDS PAGE gel and blots were probed with the chimeric rabbit version of SAIC-28B-8 (NBP3-20097) at 2 ug/ml before detection using an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence.

Flow Cytometry: hHR23b Antibody (SAIC-28B-8) - Azide and BSA Free [NBP3-20097] - Paraformaldehyde fixed HeLa cells permeabilized with 0.5% Triton were stained with anti-unknown specificity antibody (isotype control, black line) or the rabbit IgG version of SAIC-28B-8 (NBP3-20097, blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor(R) 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.

Background: hHR23b

hHR23b is encoded by this gene is one of two human homologs of Saccharomyces cerevisiae Rad23, a protein involved in the nucleotide excision repair (NER). This protein was found to be a component of the protein complex that specifically complements the NER defect of xeroderma pigmentosum group C (XP-c) cell extracts in vitro. This protein was also shown to interact with, and elevate the nucleotide excision activity of 3-methyladenine-DNA glycosylase (MPG), which suggested a role in DNA damage recognition in base excision repair. This protein contains an N-terminal ubiquitin-like domain, which was reported to interact with 26S proteasome, and thus this protein may be involved in the ubiquitin mediated proteolytic pathway in cells.
