Immunohistochemistry-Paraffin: Histone H1.2 Antibody (JU43-48) [NBP2-75932] - Immunohistochemical analysis of paraffin-embedded human lung carcinoma tissue using anti-Histone H1.2 antibody. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 8.0-8.4) for 20 minutes.The tissues were blocked in 5% BSA for 30 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (1/50) for 30 minutes at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Background: Histone H1.2

Histones are nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber as they play a critical role in transcription regulation, DNA repair, DNA replication, and chromosomal stability. The Histone H1.2 protein is 213 amino acids long, 21 kDA and is encoded by the HIST1H1C gene. H1 proteins are essential in forming chromatin fiber and for the condensation of nucleosome chains into higher-order structured fibers. The HIST1H1C gene has been investigated in leukemia and parkinson's disease. It plays a role in apoptosis, PKA signaling, cell cycle chromosome condensation in prometaphase, and histone modification through interactions with various genes such as TONSL, NPM1, YWHAQ, EED, and ESR2.
