Background: Complement Component C3aR

Complement Component 3a receptor (C3a R) is a 70-80 kDa G-protein coupled 7-transmembrane sulfated glycoprotein that belongs to the G-protein coupled receptor 1 family of proteins. It is expressed inducibly or constitutively by platelets, eosinophils, monocytes, astrocytes, CD4+ T cells, dendritic cells, NK cells, mast cells and basophils, neutrophils, adipocytes, osteoclasts, osteoblasts macrophages, vascular endothelial cells, bronchial epithelium and smooth muscle. Engagement of C3a R by  77 kDa C3a has a number of effects that are context-dependent. C3a action on adipocytes facilitates glucose and fatty acid uptake, while in bone, C3a R (plus C5aR) drive the generation of Th1 and Th17 cells. Perhaps its best action is that of a mediator of anaphylaxis. On mast cells it induces histamine release, visceral smooth muscle contraction and vasodilation. Human C3a R extracellular regions share 50% and 42% amino acid identity with mouse and rat C3a R, respectively.
