Background: FRS2
FRS2 (FGFR substrate 2; also SNT and FSR2-alpha) is a 70‑90 kDa member of the FSR family of lipid-anchored docking proteins. It is an intermediary between FGF and TRK receptors, and their Ras/MAPK signaling cascades. Human FRS2 is 512 amino acids (aa) in length. It contains a membrane-anchoring myristoylation signal (aa 1‑6), a PTB domain that interacts with FGF and NGF receptors (aa 13‑115), and a C-terminal Tyr-rich region that serves as a docking site for Grb2 and Shp2 (aa 196‑471). One splice form shows a deletion of the C-terminal four amino acids. Over aa 121‑449, human FRS2 is 99% and 94% aa identical to dog and mouse FRS2, respectively.
